چکیده

the satellite cells. Contaminating CD45+/Mac-1+/Sca-1+ cells were subsequently removed by MACS cell separation. Cell cycle analysis revealed that over 80% of the resulting myogenic cell population was in G0, indicating a high proportion of quiescent cells in the isolated population. RT-PCR analysis showed that the expression of three Kruppel-like factors (Klf) was increased in quiescent satellite cells and decreased during a time-course of activation in culture. Furthermore, this decrease in expression correlated with the increasing expression of MyoD, which is upregulated in activated satellite cells, indicating a potential role for the Klf genes in the regulation of satellite cell quiescence.

Cell death-induced regeneration in wing imaginal discs requires JNK signalling.

Regeneration and tissue repair allow damaged or lost body parts to be replaced. After injury or fragmentation of Drosophila imaginal discs, regeneration leads to the development of normal adult structures. This process is likely to involve a combination of cell rearrangement and compensatory proliferation. However, the detailed mechanisms underlying these processes are poorly understood. We hav...

ROS-Induced JNK and p38 Signaling Is Required for Unpaired Cytokine Activation during Drosophila Regeneration

Upon apoptotic stimuli, epithelial cells compensate the gaps left by dead cells by activating proliferation. This has led to the proposal that dying cells signal to surrounding living cells to maintain homeostasis. Although the nature of these signals is not clear, reactive oxygen species (ROS) could act as a signaling mechanism as they can trigger pro-inflammatory responses to protect epitheli...

Analysis of the Function of Apoptosis during Imaginal Wing Disc Regeneration in Drosophila melanogaster

Regeneration is the ability that allows organisms to replace missing organs or lost tissue after injuries. This ability requires the coordinated activity of different cellular processes, including programmed cell death. Apoptosis plays a key role as a source of signals necessary for regeneration in different organisms. The imaginal discs of Drosophila melanogaster provide a particularly well-ch...

Gene expression during imaginal disc regeneration detected using enhancer-sensitive P-elements.

When imaginal disc fragments from Drosophila are cultured in adult female hosts, they either duplicate the part of the pattern specified by the fate map, or regenerate to replace the missing part. The new tissue is added by proliferation of a small number of cells from the cut edge, brought together when the wound heals to form a regeneration blastema. Specification of the new pattern has been ...

Drosophila Wnt and STAT Define Apoptosis-Resistant Epithelial Cells for Tissue Regeneration after Irradiation

Drosophila melanogaster larvae irradiated with doses of ionizing radiation (IR) that kill about half of the cells in larval imaginal discs still develop into viable adults. How surviving cells compensate for IR-induced cell death to produce organs of normal size and appearance remains an active area of investigation. We have identified a subpopulation of cells within the continuous epithelium o...
